In a radiant-in-slab heating system, would it be best to try and eliminate the majority of metallic components?

Ex: PEX with EP fittings and CPVC transitions to pumps etc. properly stabilized.

Also, I'm familiar with 02 migration. Barrier or no barrier?

Use PEX-AL-PEX for hydronic systems :D

Yes, no joints and no metal in ground is the only way to do the job.

depending the slab size and heat output required will affect your layout and loop spacing many times we done 8-10" loop spacing, sometimes we cheat it for 1" sometimes to fit the loops inside doorways or on the way back into the mechanical room where the manifold is, so what this would mean in get a larger spool, and if possible get an engineer to layout your piping for optimum heat

Radiant, use oxy barrier or heat pex. Metallic fittings should not be installed below the slab but for that matter, there shouldn't be any fittings below or in the slab anyway. Once you get above the concrete you can use whatever you want, it will make no difference. Most manifolds are copper or brass as are control valves, etc.
